OBJECTIVES/GOALS: We aim to discover safer and more effective therapeutics for CNS disorders. Current therapeutic development is hindered by dosing out drugs for safe consumption. By identifying proteins with narrow functional roles in the brain (i.e., behavioral control), we can develop drugs targeting these proteins for improved treatment safety and efficacy. METHODS/STUDY POPULATION: We focused on an evolutionarily new, non-neuronal, non-synaptic glutamate signaling mechanism, system xc- (Sxc). Sxc activity was eliminated by mutating the gene Slc7a11 through pronuclear injection of zinc-finger nucleases into Sprague Dawley rat embryos to create a line of rats lacking Sxc (MSxc). To confirm Sxc mutation, we verified that tissue from MSxc rats had a complete lack of xCT, which is the regulatory subunit of Sxc that is encoded by Slc7a11. We also verified that astrocyte cultures generated from MSxc tissue lacked cystine-evoked glutamate release. Next, we measured development (body weight), CNS regulation of metabolism, and other indicators of generalized, non-specific brain function as well as behaviors that are reliant on behavioral control, such as impulse control and response inhibition. RESULTS/ANTICIPATED RESULTS: Eliminating Sxc was not lethal and did not impair development or produce widespread changes in brain function as is commonly observed when deleting other glutamate mechanisms. MSxc rats did not differ from wildtype in growth rate, central regulation of metabolism as reflected by absolute or diurnal changes in core body temperature, locomotor activity in a familiar or novel environment, or simple forms of cognition such as novel object recognition, or operant responding (food and cocaine-reinforced). In contrast, behaviors that rely on behavioral control were impaired. MSxc rats displayed deficits in impulse control and behavioral flexibility. We hypothesize that MSxc rats will also show deficits in response inhibition using the stop signal reaction time task, a common metric used in clinical populations. DISCUSSION/SIGNIFICANCE: Eliminating Sxc activity in rats produced deficits in behaviors reliant on impulse control, without impacting development or simple brain function. These results show the potential of targeting Sxc to restore behavioral control without generating therapeutically limiting adverse effects resulting from non-specific changes in brain function.

These results highlight the potential of targeting Sxc to enhance cognition without generating therapeutically limiting adverse effects resulting from non-specific changes in brain function.
GALEX UV Catalog of Low-redshift Galaxies for Estimating Transient Rates

Detection of transients such as supernovae (SNe) and kilonovae (KNe) in early phase has recently become important for understanding the progenitor properties and multi-messenger astronomy. Predicting which galaxy has the higher probability of hosting the transient events would help detect the early phase of the events and get information on their progenitors. The SN and KN rates are known to be a function of star formation rate (SFR) and stellar mass of the host galaxy. The SFR of a galaxy can be estimated from ultraviolet (UV) luminosity. However, the UV magnitudes have been derived carefully only for a limited number of nearby galaxies. Here, we introduce GALEX galaxy catalog of all-sky UV brightness of low redshift galaxies. To do so, we derive the UV photometry of galaxies in the GLADE catalog using the GALEX AIS images, supplemented by GALEX NGS and MIS data. From the near-UV (NUV) and far-UV (FUV) magnitudes, we calculate the SFRs of the galaxies, which will further be useful for estimating the SN and KN rate. The results are compared with previous GALEX UV catalog of galaxies. There will be an updated catalog based on this catalog for calculating KN rate of the galaxies in the future work.

Summary
Conceptually, extending the premise of bioelectronic interfaces down to the scale of single molecules is a straightforward goal. In practice, the challenges are purely technological. Single-molecule bioelectronic devices would have to involve features much smaller than state-of-the-art semiconductor electronics, and successful design would have unique requirements for sensitivity and stability.
These imposing specifications are balanced by the potential of enormous rewards, because single-molecule bioelectronics would be a breakthrough technology for biochemical research and applications. By peering past the ensemble behaviors of traditional characterization, single-molecule techniques aim to directly observe the stochastic fluctuations, instantaneous dynamics, and non-equilibrium behaviors that make up a molecule’s full functionality. Moreover, single-molecule measurements can uncover the unusual reaction trajectories of a genetically mutated protein or a receptor interacting with pharmacological inhibitors. Building a better understanding of the precise roles of proteins in complex biological processes is a grand challenge for biology, biochemistry, and biophysics in the twenty-first century.
These potential benefits have spurred the development of a variety of single-molecule techniques. Single-molecule fluorescence, specifically Förster resonance energy transfer (FRET), has become a standard tool for single-molecule biochemistry [1]. Meanwhile, single-molecule bioelectronics has remained elusive, despite the wide-ranging capabilities of modern solid state electronics.

Disposable Smart Plastic Biochips For Clinical Diagnostics

This paper presents an overview of the development of novel disposable smart plastic fluidic biochips for clinical diagnostic applications. The biochip is manufactured using a low-cost, rapid turn around injection molding/embossing process on a plastic substrate. The plastic fluidic biochip uses a novel sPROMs (structurally programmable microfluidic system) approach to achieve passive control of fluidic sequencing [1-2]. The plastic biochip also uses an on-chip pressurized air source for fluidic movement thus eliminating the need for active driving mechanisms and allowing for a truly disposable approach. Furthermore, electrochemical biosensors are also integrated on-chip to analyze various metabolically significant parameters such as PO2(partial pressure of oxygen), Glucose, Lactate,and pH. The fluidic biochip is being developed for point-of-care health monitoring applications where parameters such as small size, simplicity of operation, disposability, reduced cross-contamination are vital. The issues mentioned above are successfully addressed using the approach of this work and are discussed in this paper.
